This study investigates the lipopolysaccharide (LPS) of the deep rough mutant I-69 Rd¯/b+ of Haemophilus influenzae. Two oligosaccharides were isolated from de-O-acylated LPS and their structures were determined using NMR spectroscopy. These structures were identified as αKdo-4P-(2→6)-βGlcN-4P-(1→6)-αGlcN-1P and αKdo-5P-(2→6)-βGlcN-4P-(1→6)-αGlcN-1P. The study also confirmed the specificity of existing monoclonal antibodies (mAbs) and assigned the epitope specificity of a new mAb (S42-10-8) to an epitope composed of the bisphosphorylated glucosamine backbone of lipid A and Kdo-4P, with the latter dictating the specificity based on phosphate group position.
